Description: Describes tests of the one geneone enzyme hypothesis involving trytophan synthetase and also adenylosuccinase. Gives an example of the use of neurospora for identifying biosynthetic pathways (in tryptophan synthesis). Presents studies of the genetic determination of protein structure (the work with hemoglobin is described in detail) which led to restating the hypothesis as one gene (cistron), one polypeptide chain. Reviews the evidence accrued from studies of the mutability of monochromatic ultra-violet light, studies of bacterial transformation by DNA, and studies with radioactive bacterial virus which indicate that DNA is the true genetic material. Lecture given by Dr. G. W. Beadle.
